Production of human motor neurons from stem cells is gaining speed

The motor neurons that innervate muscle fibres are essential for motor activity. Their degeneration in many diseases causes paralysis and often death among patients. Researchers at the Institute for Stem Cell Therapy and Exploration of Monogenic Diseases (I-Stem – Inserm/AFM/UEVE), in collaboration with CNRS and Paris Descartes University, have recently developed a new approach to better control the differentiation of human pluripotent stem cells, and thus produce different populations of motor neurons from these cells in only 14 days. This discovery, published in Nature Biotechnology, will make it possible to expand the production process for these neurons, leading to more rapid progress in understanding diseases of the motor system, such as infantile spinal amyotrophy or amyotrophic lateral sclerosis (ALS).

New genomic editing methods produce better disease models from patient-derived iPSCs

Highly valuable for modeling human diseases and discovering novel drugs and cell-based therapies, induced pluripotent stem cells (iPSCs) are created by reprogramming an adult cell from a patient to obtain patient-specific stem cells. Due to genetic variation, however, iPSCs may differ from a patient’s diseased cells, and researchers are now applying new and emerging genomic editing tools to human disease modeling, as described in a comprehensive Review article published in Stem Cells and Development, a peer-reviewed journal from Mary Ann Liebert, Inc., publishers. New method for reducing tumorigenicity in induced pluripotent stem-cell based therapies

The potential for clinical use of induced pluripotent stem cell (iPSC) technology for transplant-based therapeutic strategies has previously been hindered by the risk of dysregulated cell growth, specifically the development of tumors. The ability to use etoposide treatment to halt teratoma formation in iPSCs for the treatment of heart disease, specifically acute myocardial infarction, is demonstrated in an article in Stem Cells and Development, a peer-reviewed journal from Mary Ann Liebert, Inc., publishers. Does intravenous transplantation of BMSCs promote neural regeneration after TBI?

The brain has a low renewable capacity for self-repair and generation of new functional neurons in the treatment of trauma, inflammation and cerebral diseases. Cytotherapy is one option to regenerate central nervous system that aim at replacing the functional depleted cells due to traumatic brain injury (TBI). Bone marrow mesenchymal stem cells (BMSCs) are also considered a candidate for cytotherapy because they can differentiate into neurons/nerve cells, pass across blood-brain barrier, migrate into the injured region, secrete neurotrophic factor, and provide microenvironment for neural regeneration. Prof. Mohammad Ali Khalili, Research and Clinical Center for Infertility, Shahid Sadoughi University of Medical Sciences, Iran and his team administered TBI rats 3×106 BMSCs via the tail vein and found that the BMSCs transplanted via the tail vein promoted nerve cell regeneration in injured cerebral cortex, which supplement the lost nerve cells. Related results were published in Neural Regeneration Research (Vol. 9, No. 9, 2014).

Inner ear stem cells hold promise for restoring hearing

Spiral ganglion cells are essential for hearing and their irreversible degeneration in the inner ear is common in most types of hearing loss. Adult spiral ganglion cells are not able to regenerate. However, new evidence in a mouse model shows that spiral ganglion stem cells present in the inner ear are capable of self-renewal and can be grown and induced to differentiate into mature spiral ganglion cells as well as neurons and glial cells, as described in an article inBioResearch Open Access, a peer-reviewed journal from Mary Ann Liebert, Inc., publishers. Metastasis stem cells in the blood of breast cancer patients discovered

Individual cancer cells that break away from the original tumor and circulate through the blood stream are considered responsible for the development of metastases. These dreaded secondary tumors are the main cause of cancer-related deaths. Circulating tumor cells (CTCs) detectable in a patient’s blood are associated with a poorer prognosis. However, up until now, experimental evidence was lacking as to whether the “stem cell” of metastasis is found among CTCs.
